It is unclear whether mesenchymal stem cell-derived skeletal myogenic cells contribute to skeletal muscle regeneration after muscle injury. We investigated whether mesenchymal stem cells can differentiate into skeletal myogenic cells by following the fate of mesenchymal stem cells that express Pax7 and co-express with MyoD after muscle injury. In neonatal mice, we demonstrated that MyoD-positive skeletal myogenic cells were generated from Pax7-positive mesenchymal stem cells. Pax7-positive mesenchymal stem cells were also present in adult mice, but MyoD-positive skeletal myogenic cells were not present in skeletal muscle, suggesting that they are derived from bone marrow-derived cells. We further investigated the in vitro and in vivo fate of bone marrow-derived skeletal myogenic cells. Bone marrow cells (as sources of myogenic cells) expressed stem cell antigen-1 (Sca-1) and did not express sarcomeric myosin. After transplantation into injured muscle, bone marrow-derived Sca-1-positive cells migrated to damaged muscles and expressed Pax7. Interestingly, some Pax7-positive cells had weak MyoD expression, suggesting that some of the skeletal myogenic cells in injured muscle are derived from bone marrow-derived cells. These data indicate that the balance between bone marrow-derived and mesenchymal stem cell-derived skeletal myogenic cells is dynamic after muscle injury and that mesenchymal stem cells may be the source of skeletal myogenic cells in adults.Vignette
